Successful ligation requires relatively low temperature to achieve satisfactory annealing between two DNA fragments prior to its ligation. However, the relatively low temperature (16 oC) slows down T4-Ligase activity. "BIOGASE - fast ligation kit" contains a unique buffer and extremely active T4-Ligase-BIOGASE, which enables efficient annealing even at room temperature. Thus, ligation is completed in a few minutes.
